Ruby was a human adult during The Plague aka Black Death aka the bubonic plague and living in somewhere in Western Europe, where is never specified. At some point during her human life, Ruby started practicing magic. In order to succeed at her craft, she sold her soul to a demon named Tami. Sometime after making her deal, Ruby contracted the plague and died. According to the deal she’d made for more magic Ruby’s soul was dragged straight to Hell.
Very little is actually known about Ruby and much of her history is open to speculation. However, she most assuredly spent a great deal of time in Hell. At some point during her stay, she made the transition from tortured soul to demon. She once tells Dean that Hell burns away your humanity, it burns away your memories but for reasons that have never been revealed, Ruby isn’t like most demons. She remembers what it was like to be human and perhaps holds onto some shred of her humanity because of this.
Whether Ruby has spent the entire time since her death in Hell or not isn’t clear. One can assume she’s been above ground at least a few times in the last several centuries because she fits in. She uses slang and more current language, dresses appropriately and appears to be comfortable with this century.
Ruby gets her big break out of Hell when the hunters, Sam and Dean Winchester, open the gates of hell and let a flood of demons out. She is one of many and like some of the others she shadows Dean and Sam, waiting for the right time to make her move. The right time comes in the form of Sam needing a savior. She fills in, saves his ass, and takes off without ever telling the youngest Winchester who she is. Lucky for Sam, and Ruby as well, she has a knife; the only one in existence that can kill demons; not just send them back to hell. In Ruby’s words “Comes in handy when you’re saving the damsel in distress.”
In Hell, topics of conversation are limited. There’s who’s new, who’s screaming, and Sam Winchester. Sam is prophesied to be the Anti-Christ and Ruby is betting on that horse. She wants Sam to defeat Lilith and lead an army to keep demons from destroying the world because Ruby likes the world; primarily because it's not Hell and she'd like a permanent vacation from that place. If he does this, he'll be the King of Hell, since it's really not Sam's sort of gig, Ruby will be kind enough to step in and be Queen so Sam can go back to doing his cute little savior thing. In order to get Sam to do this, Ruby has to gain his trust. Saving him is just the first step. The second step is a lie. Sam’s brother, Dean, sold his soul to the devil and in return he got a year to live. Sam is desperate to save Dean and Ruby plays upon that. She tells Sam that she can help him save Dean but that's a lie. Dean is beyond help but she’s a demon and lying goes with the territory. There is a gun called The Colt. It was built by Samuel Colt the night all those men died at the Alamo. This gun can kill demons; in fact, this gun can kill anything it hits as long as the shot is fatal [IE heart or head—with the exception of angels as we find out later in the series. I suspect, though, that not even Ruby knows that]. The Winchesters have the Colt but the problem with it is that Samuel Colt only made 13 bullets to go with and he put all the magic in the bullets. The Winchesters have run out of bullets, which is where Ruby comes in. She helps fix the gun so that all the magic is in the gun and any bullet put in it will be just as deadly as the original 13. Of course, in the process she gets shot, but it’s a minor detail as gunshot wounds only make nasty scars. They don’t actually kill a demon provided the gun wasn’t the Colt. The Colt is an integral part of Ruby’s plan as demons actually are afraid of things that can really kill them.
By this time, Sam has bought almost whole hog into Ruby’s story. Her fourth act of trust comes in saving Dean from a witch’s hex. She’s off just as quickly to save Sam and Dean from the same witch that hexed Dean previously. Ruby is ready at this moment to sacrifice herself for Sam and, by proxy, Dean. She nearly makes that sacrifice, but Dean ends up being the one to save her.
The Winchesters are never out of trouble for long. Shortly after this they end up in jail and the jail, naturally, ends up surrounded by demons that want to kill Sam. It's Ruby to the rescue again, or so the plan goes. She can’t let them kill her own personal Jesus Christ. She fights her way through the hoard of demons, gets in the jail and offers up a plan that Dean violently protests to. She knows a spell to kill all demons in the area, including herself but it requires the heart of a virgin. Lucky they have one on hand. Sam is ready to go along with plan, liking the idea of killing one to save many. Dean protests , convinces Sam and Ruby leaves thoroughly disgusted. She tells Sam that she bet on the wrong pony (him over Lilith). This is the point (in my opinion) where Ruby gives up on Sam and switches teams. Before Sam 'betrays' (in her eyes) her she's been absolutely truthful about what she wants. Sam is her little anti-Christ and she wants nothing more then to see him stop Lilith from making the world Hell on earth. After all, the world has french fries, ketchup and tall men. She steps outside the door of the jail, asks if anyone wants to fight her and the sea of demons parts to let her pass. In the end, Ruby’s plan was the better one. Sam and Dean make it out but Lilith comes in shortly after and destroys the jail along with everyone in it. She bursts into Sam and Dean’s room, makes them watch the news and tells them ‘Next time, we do my plan’.
The last time we see Ruby is a few weeks after this, the day Dean’s deal comes due. Sam summons her and asks for her knife in order to kill Lilith. That’s not Ruby’s plan and she doesn’t hand over the knife willingly. This results in a fight between Dean and Ruby. In the end, Ruby ends up locked in a devil’s trap and Dean has the knife. After some time, Ruby escapes the devil’s trap and tracks the boys down. She meets them in New Harmony, Indiana where Lilith has been ‘vacationing’. At some point after Ruby shows up, Lilith leaves the little girl she’s possessing behind and possesses Ruby’s body instead, evicting Ruby and sending her back to Hell to pay for her sins. The Hellhounds come to get Dean, Ruby/Lilith opens the door to let them in, pinning Sam to a wall so he has to watch his brother die. The last we see of Ruby, Lilith has vacated the body and left a corpse behind.**

Personality:
Ruby is a bitch. She’s sarcastic, condescending and insulting. Her social skills are nil to nothing. In part, it's a coping mechanism to keep humans at a distance. Ranchers don't get attached to the cattle but it's also just who she is. As a demon she doesn't look at the world, people, emotions or social graces the same way. She's not capable of some of the emotions that humans have and even if she were, she doesn't see the point. Humans are beneath her: nice, cozy bodies to burrow into and entertainment on a Friday night. She’s selfish and self-serving, going to almost any lengths to save her own skin. Ruby would sell out her own mother to save her skin and everyone knows it. This is motivated partially by fear. Ruby doesn't want to go back to Hell. Crawling out was no easy feat and she knows if she returns there will be hell to pay for the things she's done topside. Her selfishness and self-serving attitude are also founded on the fact that Ruby likes herself better than she likes just about anyone else. She feels a little superior to other demons because she has retained a shred of her humanity and she feels a lot superior to humans because she's not human.
Ruby enjoys causing other people and demons to squirm. It's a basic bully mentality and is probably derived both from her human life and her existence as a demon. She gets absolute glee out of making people uncomfortable. As a human and a witch, it's likely she enjoyed causing people who 'deserved' it to squirm as well and as a demon that's carried over. She’s smart as a whip and vicious to boot, willing to go that extra mile to get things done and by extra mile I mean of course slaughtering a mile of whatever she has to in order to get the job done. She doesn't back down from doing the hard things. Sure, as a demon the hard things don't bother her much, but she's also learned that the hard things have to be done and no one lines up to do those.
She is also quite manipulative, knowing when to back down, play sweet and innocent when in reality she’s neither. Even if she wanted to be, her basic nature won't allow sweetness or innocence. The most she can manage is to not be a bitch. Duplicity is a skill she's had to learn over the centuries in order to survive. She manipulates Tami into believing that she's ready to come back to the fold with a little seduction and some charm. She knows exactly which of Sam's buttons to push in order to gain his trust and she even manages to make some headway with Dean.
For all of Ruby’s cynicism and bitchiness, she loves living on earth, as opposed to being stuck in Hell. We can see some of this in the way she eats her french fries with gusto and slurps down her shake in the same manner. She doesn't have to eat as a demon but she enjoys it. Because of the many years she did spend in hell, she appreciates the little things in life. She is, at heart, a demon though. She doesn’t squelch from blood or pain or combat. She’s a little fighting machine (after all one doesn’t just walk out of Hell) and she’s brutally practical about things. For a demon, she’s even kind of loyal (which isn’t honestly saying much). She sticks by Sam though a lot of failures, placing her bets on a pony that isn't slated to win this thing because she's not happy with the status quo. Granted, she does finally give up and betray him but it's at a much later date than anyone placing bets would have estimated.
There's some speculation on whether Ruby was pulling a scam all along or whether she was telling Sam the truth throughout most of season 3, I think it's a matter of interpretation. In the majority of S3 we see Ruby being fairly loyal to Sam, even risking her life in order to save him. We know there is a 'Team Sam' because of something Casey, the demon from Sin City, says. She tells Dean that she was ready to throw in with Sam and she wasn't the only one. This is a hold over from Azazel's plan's for Sam and the 'Special Kids' but I think Ruby is taking up the mantel of this in order to save her skin and get rid of Lilith. It puts her in a good position. If Sam succeeds, he ends up being the defacto leader of Hell while Lucifer is locked up in a cage—a position that Crowley has taken up. One could assume Sam wouldn't want this position so he would hand it off to the most likely candidate, Ruby. That puts her in a very, very good position. The earth remains intact, Hell is hers to do as she sees fit and she's the Queen of Hell. All of this only pans out if Sam wins.
It's important to note that Ruby says a couple of times in S3 that she doesn't believe in the devil. She doesn't say she doesn't worship him or he's not her god but that she doesn't believe in him.
Jus In Bello is a very important episode for S3 & S4 Ruby. I mentioned that she feels Sam betrays her by not killing the virgin and wiping out Lilith. It's a turning point and a test for Sam, to see just how far he's willing to go. He fails spectacularly and it becomes clear, in Ruby's mind, that Sam isn't going to win this fight with Lilith. He's got the potential but he doesn't have the stones. She's forced to make a decision, stay with the losing horse and hope she can pull out an 11th hour miracle or throw in with the favorite, Lilith. The moment she steps out of that jail, she's on Lilith's side. I think this is shown very well in the episode by the fact that Ruby had to fight her way INTO the jail but when she leaves, the waters part and the demons let her walk on by. Canonically we don't know what happened between the time Ruby left the jail and the time she showed up in the Winchester's hotel room at the end of the episode but I believe that she took this time to make a deal with Lilith: She would bring the Winchesters to her and then once Dean is out of the way, she will make sure Sam is ready to not only help break the seals and release Lucifer but to be Lucifer's vessel. In return for this, Ruby would get to be Lucifer's right hand man.
This sets up the way s4 Ruby behaves. She plays the weaker, scared, more defensive part because that's what Sam needs her to be. It will activate that white knight complex and he will step up to the plate. At least that's what she hopes. It is also the part that Lilith needs her to play. In order to keep up their end of the deal and come out of this smelling like a rose, Ruby has to be submissive to Lilith. She's screwed up, she's been branded as a traitor and now she's got a year to prove where her loyalty lies. Ruby can be devoted but only as far as it benefits her. In s3 her loyalty was with Sam because she thought he could help her. In s4 her loyalty is to Lilith because she will be her saving grace.
In the episode Malleus Maleficarum, it can be assumed from Ruby's conversation with Tami that she's pulled something very much like this before. Betraying her loyalties for something better isn't anything new to Ruby.

Abilities, Strengths and Weaknesses:
Ruby has supernatural strength and advanced hearing plus the ability to detect demons, angels and other supernatural things. She also has telekinesis, teleportation, some advanced healing, inhuman durability and the ability to possess different bodies, but she’s pretty attached to the blonde she’s got so possessing another body would only be something she’d do in dire circumstances.
She's also a kick ass fighter as she should be. She's been doing it for hundreds of years. She can do spells as long as she has the components (think witch stuff, not inborn magic stuff) and has a vast supernatural knowledge.
Outside of this, Ruby is a mess of strengths and weaknesses. I’ve addressed a few of these in her personality, but I’ll reiterate them here. She’s a bit unclear on the definition of loyalty. While she’s willing to bust her cute, little, demon ass to make things work out in her favor, but she’s not willing to fail. She’s got certain clear goals, but she’ll sacrifice those goals for her own skin. She’s not really a team player, preferring to work on her own in her own way. She’s not good at being social or nice or human.
On one hand, Ruby prefers the position of leader, but she’s a shit leader. She wants things done her way, but she prefers blind faith to informed following. She cares more than she wants to, but not enough to make a difference in most of her actions and behaviors. She's sort of stuck between not as human as she needs to be nor as demon as she needs to be. She doesn't care about the human race as a whole, but she wants to preserve humanity mostly because she likes the things that humanity brings to the world at large.

“God dammit, Sam, do you even want to save Dean?” Ruby is standing in the middle of the room, a look of pure disgust on her face. She looks bored as well as disgusted and she examines her fingernails blandly. She’s prepared for the backlash, the anger and even the brink of tears her question pushes him to.
“Don’t.” It’s just one word but he makes it sound dangerous in a way that sends chills up and down her spine. Not that anyone could tell that from looking at her. She does love a tall, bad boy.
“Don’t what, Sam? You’ll hit me? Please. You hit like a girl.”
One big wailing punch and Ruby hits the ground, hands going behind her to keep her back from hitting the floor. She just laughs. The more she pisses him off, the better this works for her. Eventually he’ll figure it out on a whim but for now its okay to use anger as a crutch.
“SHUT UP!” It’s a bellow more than anything so filled with pain, frustration and anger that the words are nearly incomprehensible. She knows he’s trying. He’s just not trying hard enough.
“Make. Me.” Each word is enunciated, turning them into a dare and a challenge that she’s hoping Sam won’t pass up. She watches while he falls apart, hands going over his ears and if Ruby believed in any sort of deity she might pray that she can put him back together. In fact, she’s counting on it. This is one time in her existence that Ruby has to be right because if she’s not, the whole world is going to pay.
“Don’t ignore me, Sam. I’m not your dying brother. You remember Dean, the guy who is afraid of going to Hell? The guy who was counting on your lazy ass to save him? Didn’t you promise him that you’d save him? Talk about faith misplaced.”
He degrades before her eyes. It’s like watching someone torture a puppy, her puppy. He won’t believe her—no one would—but she hates this part of the job. It would be so much easier to go from hunter to General with a snap of her fingers but that’s not the point. That’s how Azazel would do this and she needs Sam to still be Sam when he gets to the end of the road.
“STOP IT!” Sam yells and this time she’s flung from the floor and slammed against the wall but Sam never moved a muscle. Sam never twitched a finger. Almost as soon as she hits the wall, she falls back to the floor but she’s up on her feet and she’s clapping. It’s a slow walk toward him and her arms are around his neck, lips pressed to his.
“That’s my little Anti-Christ,” she smirks when she pulls away from him. Puppy’s going to make it yet.
